Özet

Arrhythmogenic properties of catecholamines become evident in the presence of inhalational agents. The effects of midazolam and propofol in this respect have not been compared before. The effects of midazolam and propofol on adrenaline-induced arrhythmias were compared in rats. After approval from the Ethics Committe of Research of Laboratory Animals, the study was conducted in three stages in 50 rats. In the first stage, ED50 dose of midazolam infusion was determined in 20 rats. In the second stage, the sensitivity of 30 male rats to adrenaline was investigated under halothane anesthesia. In the third stage, midazolam (ED50) and propofol (ED50, 39 mg kg-1 hr-1) were infused to sensitive male rats for an hr in a randomized manner. Thereafter, the doses of adrenaline that caused arrhythmias were determined. Analysis of logistic regression, Kruskal-Wallis and Mann-Whitney U tests were used. ED50 dose of midazolam infusion was determined as 9 mg kg-1 hr-1. The mean dose of adrenaline that caused arrhythmia in rats which were sensitive to the drug under halothane anesthesia (n=16, 5.8±0.6 μg kg-1) was significantly lower than both the mean doses of adrenaline that caused arrhythmia after midazolam (n=8, 68.0±19.2 μg kg-1) and propofol infusion (n=8, 102.0±17.0 μg kg-1), (p<0.001, p<0.001, respectively). There was no significant difference between midazolam and propofol groups in this respect (p=0.195). It is concluded that both midazolam and propofol have less effect than halothane on ventricular arrhythmogenic properties of adrenaline. Additionally, midazolam, and propofol have similar effects on adrenaline-induced arrhythmias in rats.
