Ser159 mutation of the pre-initiation complex protein TAF7 CAUSES A G2/M block in cell cycle

Abstract
Objective: TATA Binding Protein (TBP) associated factors (TAFs) have been shown to participate in transcriptional regulation. TAF7 is one of the TAFs, which role has been emphasized in the regulation of transcription and cell cycle. It has been suggested that TAF7 acts at a check point at the initiation of transcription by not leaving the pre-initiation complex until it is completely and correctly built. It has also been reported that TAF7 play a role in cell cycle regulation and is hyper-phosphorylated on Ser159 at the G2/M checkpoint. To study the effect of this phosphorylation on cell cycle kinetics, we did cell cycle analyses on WT and TAF7 Ser159 mutant cells in a tetracycline controlled expression system.
