Effects of Apilarnil on Type 2 Diabetes-Induced IRS-1/PI3K/Akt Mediated Insulin Resistance in Male Rats

Diabetes is one of the most important chronic and metabolichealthproblemsseenasanepidemicof the21stcentury.The incidenceof diabetes is gradually increasing, anddifferentmethods are usedtotreat it.Apitherapyproducts suchashoney, propolis, royal jelly, andbeevenomcanbeusedasanalternativetotreatdiabetesandreduce symptoms.Oneof theseapitherapyproducts isApilarnil,whichhasnot yet become popular. The hypolipidemic, hepatoprotective, androgenic, anabolic, and immunesystemenhancingpropertiesofApilarnil indicate that itmaybeeffectiveinthetreatmentof type2diabetesor inreducing the symptoms.The studyaimed todetermine the therapeutic effect of Apilarniloninsulinresistanceandthepossibleunderlyingmechanism. In thestudy,408week-oldmaleWistaralbinoratswereused.Theratswere dividedinto5groupsasControl,Diabetes,Diabetes+Api1(125mg/kg/ daylyophilizedApilarnil),Diabetes+Api2(250mg/kg/daylyophilizedApilarnil)andDiabetes+Api3(500mg/kg/daylyophilized Apilarnil).Eachgroupconsistedof8rats.Apilarnildoseswereadministeredbyoralgavagewith1mLofdistilledwater,5daysina week, during the last6weeks.The levelsofNrf2,NF-κB,TNF-α, total IRS-1, p-IRS-1, PI3Kp85, totalAkt, p-Akt, IL-1B, and GLUT4proteinsweredeterminedintargettissuesbyWesternblotting.Alanineaminotransferase(ALT),aspartateaminotransferase (AST),high-densitylipoprotein(HDL), low-densitylipoprotein(LDL), total cholesterol (TC), andtriglyceride(TG)levelswere detectedintheserum. Serumsuperoxidedismutase(SOD), catalase(CAT), glutathioneperoxidase(GSH-Px),malondialdehyde (MDA),andinsulinlevelsweredeterminedbytheELISAmethod.Accordingtothedataobtainedinthestudy, theapplicationof Apilarnil, comparedtothediabetesgroup,balancedsomefindings,butdidnotdemonstratea fullyantidiabeticeffect.
